چکیده
Embryonic wound healing is both a mechanical process, driven by intraand intercellular forces, and a genetic process, regulated by an array of signalling molecules. Our goal is to understand the fundamental biology and dynamics of embryonic wound healing – to connect the phenomenology of wound-margin shortening to the underlying cytoskeletal mechanics and genetic regulatory networks. As a model system, we are using the epidermal tissue of fruit fly embryos (Drosophila melanogaster) at stage 15, just after formation of the epidermis. This system is well-suited to integrated genetic and mechanical studies because we can generate precise wounds with laser-microsurgery, and follow the cellular details of wound closure in living, GFP-labeled embryos, both in wild type and in mutants, via confocal microscopy. We are collaborating with a group of computational biologists to generate a mathematical model of wound healing that will help us to understand this process.

Leukocyte Tyrosine Kinase Functions in Pigment Cell Development
A fundamental problem in developmental biology concerns how multipotent precursors choose specific fates. Neural crest cells (NCCs) are multipotent, yet the mechanisms driving specific fate choices remain incompletely understood. Sox10 is required for specification of neural cells and melanocytes from NCCs. Like sox10 mutants, zebrafish shady mutants lack iridophores; we have proposed that sox1...
متن کاملALKALs are in vivo ligands for ALK family receptor tyrosine kinases in the neural crest and derived cells
Mutations in anaplastic lymphoma kinase (ALK) are implicated in somatic and familial neuroblastoma, a pediatric tumor of neural crest-derived tissues. Recently, biochemical analyses have identified secreted small ALKAL proteins (FAM150, AUG) as potential ligands for human ALK and the related leukocyte tyrosine kinase (LTK). In the zebrafish Danio rerio, DrLtk, which is similar to human ALK in s...
متن کاملAlk and Ltk ligands are essential for iridophore development in zebrafish mediated by the receptor tyrosine kinase Ltk.
Anaplastic lymphoma kinase (Alk) and leucocyte tyrosine kinase (Ltk) were identified as "orphan" receptor tyrosine kinases (RTKs) with oncogenic potential. Recently ALKAL1 and ALKAL2 (also named "augmentor-β" and "augmentor-α" or "FAM150A" and "FAM150B," respectively) were discovered as physiological ligands of Alk and Ltk. Here, we employ zebrafish as a model system to explore the physiologica...
متن کاملFoxd3 controls melanophore specification in the zebrafish neural crest by regulation of Mitf.
We describe a mechanistic model whereby Foxd3, a forkhead transcription factor, prevents neural crest-derived precursors from acquiring a melanophore fate. Foxd3 regulates this fate choice by repressing the mitfa promoter in a subset of neural crest cells. mitfa is only expressed in a Foxd3-negative subset of neural crest cells, and foxd3 mutants show an increase in the spatial domain of mitfa ...
متن کاملHedgehog signaling is required for cranial neural crest morphogenesis and chondrogenesis at the midline in the zebrafish skull.
Neural crest cells that form the vertebrate head skeleton migrate and interact with surrounding tissues to shape the skull, and defects in these processes underlie many human craniofacial syndromes. Signals at the midline play a crucial role in the development of the anterior neurocranium, which forms the ventral braincase and palate, and here we explore the role of Hedgehog (Hh) signaling in t...
